Topic: Regulation and Function of Mitochondrial Biogenesis in Neurons

Mitochondrial maintenance in neurons is essential for neuronal health and function. Mitochondrial dysfunction is linked to neurodegenerative disease and has been found to underlie hair cell death after aminoglycoside antibiotic treatment. However, we have little understanding of how mitochondrial populations in these metabolically active cell types are maintained. We are studying mitochondrial maintenance in neurons using zebrafish. Using a combination of genetics and live imaging, we have found a strong association between mitochondrial transport and maintenance of mitochondrial distribution, health, and function. Specifically, disrupting retrograde mitochondrial transport causes an accumulation of damaged organelles in axon terminals. Strikingly, we also observe a loss of mitochondria in the neuronal cell body due to disrupted mitochondrial biogenesis. Transcriptional analysis has revealed a strong reduction in genes associated with mitochondrial biogenesis when mitochondrial retrograde transport is disrupted. Currently, we are working to address the frequency and function of mitochondrial biogenesis in neurons to determine how this critical organelle population is maintained to support neuronal health and function.
